We are currently seeking a SCIENTIST with MOLECULAR BIOLOGY and AUTOMATED HIGH THROUGHPUT SCREENING (HTS) EXPERIENCE to work onsite at the National Institutes of Health in Bethesda, Maryland. Bethesda is 9 miles from Washington, DC.

TASKS/SERVICES. The contractor shall:
Perform a variety of growth inhibition assays against Mycobacterium tuberculosis in BSL3 lab.
Run the BioMek FX and BioMek NX automated liquid handlers and associated microplate readers in the high-throughput screening (HTS) laboratory in a BSL3 environment.
Maintain SOPs, service agreements and organization in the HTS laboratory.
Perform QC screening operations to ensure that screens perform.
Maintain the compound inventory.
Maintain screening data files.
Perform basic molecular biology techniques on E. coli and M. tuberculosis such as plasmid transformation and mutant generation.
Maintain organization and lab stocks of screening laboratory.
Make the necessary growth media for M. tuberculosis assays.
Optimize screening assays for eukaryotic cells including HepG2 and Vero cell assays.
Manage the operation of cellular cytotoxicity and mitochondrial toxicity assays.
Run reporter assays that give readout for inhibition of bacterial cell wall synthesis.
Design a Fluidigm BioMark microfluidic system as a tool to explore mechanism of action of growth inhibitors; design includes selection of genes based on literature reports of genes that are responsive to inhibition of pathways, optimizing primers sets for these genes, implementing the system using control drugs and optimizing the running of this system.
Optimize methods for RNA isolation from small culture volumes for analysis by Fluidigm arrays.
Analyze high-throughput data generated by HTS as well as Fluidigm Biomark arrays using software such as Spotfire and GeneSpring.
REQUIREMENTS. The contractor must have:
Master degree in a scientific discipline. Two (2) years of specialized experience plus a Bachelor degree is equivalent to a Master degree.
Minimum two (2) years of experience as described above in TASKS.
Experience working in a BSL3 laboratory.
Two years of experience with aseptic technique, PCR, DNA/RNA extraction and other molecular biology techniques.
Demonstrates experience working in the research laboratory environment.
Strong proficiency with computer skills including MS Office Suite.
PREFERRED: whole cell screening, screening against proteins and working with Mycobacterium tuberculosis.
